目的 观察经静脉途径移植骨髓间充质干细胞(MSCs)24小时,2周及1月后在慢性心肌梗死模型大鼠体内细胞的分布情况。
方法 从幼年雄性Sprague Dawley大鼠股骨及胫骨获取并培养MSCs。将18只雌性SD大鼠制作心肌梗死模型,3周后经心脏超声检测并将合格模型随机分为2组,第一组为实验组(n=9),将培养成熟的MSCs (5×106,300μl)通过股静脉注入模型大鼠体内。第二组为对照组(n=9),将等体积的磷酸盐缓冲溶液(PBS)经股静脉注入模型大鼠体内(n=9)。另选9只正常雌性SD大鼠,对其行开胸术,但不结扎冠状动脉,作为假手术组,并将成熟的MSCs (5×106,300μl)通过股静脉注入体内。经静脉途径移植MSCs 24小时,2周及1月后,各组大鼠体内的细胞分布情况应用实时荧光定量PCR技术(RT-PCR)进行检测。
结果 经静脉途径移植骨髓间充质干细胞24小时后,干细胞主要分布在肺组织中,实验组和假手术组细胞滞留量分别为2584269±647796和 2901384±784872, 而心脏、肝脏及脾脏中细胞分布很少。细胞移植2周后,包括肺组织在内的各个器官细胞分布均很少。细胞移植1月后,各器官中细胞数目明显减少。
结论 慢性心肌梗死后经静脉途径移植MSCs,早期大量细胞滞留于肺组织中,且细胞数目随时间延长而锐减。此外,在心脏、脾脏及肝脏中仍可有少量细胞分布。

Cell body distribution in chronic myocardial infarction rats after
bone marrow mesenchymal stem cells intravenous transplation Abstract
Objective To study the systemic distribution of bone marrow derived mesenchymal stem
cells (MSCs)in chronic myocardial infarction SD rats 24 hours,2 weeks and 1 month after cell intravenous implantation.
Methods MSCs from male Sprague Dawley(SD) rats were cultured for transplantation. Three weeks after induction of myocardial
infarction, 18 female SD rats were randomized into 2 groups. MSCs (5×106,300μl) were injected into the femoral vein of experimental group(n=9), meanwile phosphate buffered saline (PBS)of equal volume were injected into the femoral vein of control group (n=9). Another 9
female SD rats that underwent thoracotomy only were involved as sham operation group,and MSCs (5×106,300μl) were injected as the same way. The systemic distribution of MSCs were evaluated
through Real-time Polymerase Chain Reaction (RT-PCR) at time points of 24 hours, 2 weeks and 1 month after injection.
Results 24 hours after MSCs injection intravenously, cells mainly concentrated in the
lung in experimental group and sham group.The cell number was 2584269±647796
and 2901384±784872 respectively,and only few cells concentrated in the heart,spleen and liver. 2 weeks after transplantation, there were few cells in all organs
including the lung. Obvious cell loss was noted in all organs 1 month after
transplantation.
Conclusion For chronic myocardial infarction, most MSCs were early trapped in the
lung after intravenous transplantation, however, substantial number of cells lost with
prolonged time span. Only a small fraction of cells concentrated in heart, spleen, and liver.
